Abstract

Objective: This study aimed to investigate the inflammatory effects of Omp25 overexpression on BV2 cells in mice transfected with the lentivirus-mediated Omp2 gene. Brucella coli center is one of the factors which causes nervous system infection. This study is based on the study that further reveals the Omp25 gene how causes the central nervous system infection, is not only a theory, and has the practical significance, to perfect the related theory to the theory of practice through the experiment Omp25 some mechanisms of cell, discusses its influence on cells, cells to reveal the relevant pathological change process, is expected to put forward effective prevention and control measures, make a contribution to the prevention of such diseases. 

Methods: BV2 cells grown on the number of cycles were selected as the study objects. The infected LV-OMP25-GFP group was selected as the experimental group, the infected LV-GFP group was selected as the negative control group, and a blank control group was set for the study. 72h later, fluorescence microscopy was used for observation since fluorescence was labeled before the experiment, and the conversion was determined according to the observation results. At the same time, after the screening, the results should be sorted into BV2 cells expressing GFP stably, which should be completed by flow cytometry, and the operation process should be standardized and strictly abide by the laboratory rules. 

Results: The transcription expression of the Omp25 gene in the lentivirus group was significantly upregulated, and the expression was also significantly upregulated when compared with the other two groups, and the result was P<0.01. However, the results of the other two groups were meaningless, and the results of the GFP group and the blank control group were not reliable. Therefore, based on the research results, it is believed that BV2 cells successfully overexpress the Omp25 gene is correct. 

Conclusion: Due to the endocytosis of cells, Omp25 overexpressed lentivirus particles were phagocytic into BV2 cells, and the ultrastructure of the cells was greatly changed. The changes in the structure led to the unstable internal environment of the cells, and various functions were affected, which may lead to the apoptosis and death of the cells. Thus, in the NF-κB signaling pathways activated due to the overexpression of Omp25 circumstances, the cells secrete a lot of TNF alpha, IL - 6, NO; on the basis of this conclusion, Omp25 brucella bacillus disease may cause nerve type, is one of its related molecular mechanism induced, in the future research should pay attention, as a key and cannot be ignored.
