Abstract

Objective: To explore the effect of SSTR2 (somatostatinreceptor2) signal inhibition on the secretion of growth hormone in mice and its mechanism. 

Methods: SSTR2-/- mice were obtained by hybridizing the same nest SSTR2+/+ mice, and the offspring backcross parents increased the proportion of SSTR2-/- mice, and the same nest wild type was used as the control. The body size and weight of mice after birth were observed, and the expression of growth hormone protein was tested using WB test. The expression of pituitary growth hormone (GH) in mice was tested using qRT-PCR. ELISA method was applied to measure the secretion of growth-related hormones, computed tomography (CT) and bone linear growth and development difference. 

Results: The birth weight of SSTR2 knockout mice was evidently different from that of wild type mice (P<0.01). The expression of growth hormone (GH) mRNA in pituitary was down-regulated (P<0.01). GH (growth hormone) secretion in peripheral blood also decreased evidently (P<0.05). WB showed that GH expression decreased (P<0.05). CT showed that the longitudinal axis length of SSTR2-/- mice was evidently shorter than that of SSTR2+/+ (P<0.01). 

Conclusion: Knockout SSTR2 can inhibit the secretion of growth hormone in mice and inhibit the growth of mice, which is expected to become a potential therapeutic target in clinic.
